Background	Interferon-γ is a potent multifunctional cytokine which is secreted primarily by activated NK cells and T cells. Originally characterized based on anti-viral activities, IFN-γ also exerts anti-proliferative, immunoregulatory, and proinflammatory activities. IFN-γ can upregulate MHC class I and II antigen expression by antigen-presenting cells. The B27 antibody reacts with the human interferon-γ. The B27 antibody can neutralize the bioactivity of natural or recombinant IFN-γ.

Q1:组织样本检测胞内指标的注意事项有哪些？

①所有样本管均需与实验组一样进行固定破膜，如blank对照，单标组，fmo-iso；②各组固定破膜时间尽量不要相差太久，如果样本量多，建议分批次操作；③对于肿瘤组织样本，由于检测目的细胞量少，需考虑增加抗体用量，上机时间数至少100万个；④建议增加75um微米孔径滤网进行二次过滤，提高免疫细胞纯度和含量；⑤由于刺激阻断时间较长，需要在刺激阻断时间结束前注意无菌操作。

Q2:流式抗体样本什么时候用无菌的耗材？

后续需要进行细胞培养操作的实验都需要使用无菌耗材，如胞内因子检测或流式分选实验。

Q3:人外周血检测胞内因子的注意事项有哪些？

①选择肝素抗凝管；②样本制备：a.人外周血样本：提前稀释红细胞裂解液，现配现用；b.PBMC样本：将样本及试剂复温到18~20℃以后再进行实验；实验环境温度控制在18~20℃；收集的新鲜人血尽量在1 h内进行PBMC分离; 加入血液时，避免冲散分层液面；加完后不要混匀或晃动管。③采血过程中，血样在肝素抗凝管中充分摇匀，避免血凝（肝素抗凝管保存时间一般不要超过12h）；④裂红时间不超过5min，建议做预实验摸索一下最佳时间；⑤无菌操作；⑥刺激和阻断时间建议通过预实验摸索。
