| 检测原理 | 本试剂盒采用双抗体夹心ELISA法。用抗小鼠GLP-1抗体包被于酶标板上，实验时样品（或标准品）中的小鼠GLP-1会与包被抗体结合。后依次加入生物素化的抗小鼠GLP-1抗体和辣根过氧化物酶标记的亲和素，抗小鼠GLP-1抗体与结合在包被抗体上的小鼠GLP-1结合，生物素与亲和素特异性结合而形成免疫复合物，游离的成分被洗去。加入显色底物(TMB)，TMB在辣根过氧化物酶的催化下呈现蓝色，加终止液后变成黄色。用酶标仪在450 nm波长处测OD值，GLP-1浓度与OD450值之间呈正比，通过绘制标准曲线计算出样品中GLP-1的浓度。 |

Q1:E-EL-M3012 GLP-1提取用ripa的话是否有影响？
建议最好用PBS，RIPA对有些指标的检测是有影响的，尽量避免使用。不建议使用商品化的裂解试剂。不同的产品组成有差异且裂解能力也不同，如果试剂含有SDS等表面活性剂会对目的蛋白的天然构象造成影响，可能会出现测值明显降低或者假阴性。此外，由于是新引入的溶剂，不确定基质干扰的情况，可能会造成本底升高，从而影响测值准确性。
